Increasing GPT activity for colorectal cancer
Increasing activity of the metabolic enzyme GPT could help treat colorectal cancer by promoting colon production of the Krebs cycle metabolite α-ketoglutarate, which reduces tumorigenic WNT signaling, while also directly binding and promoting the degradation of the carcinogenic folate cycle enzyme MTHFD1L.
A transcriptomic analysis of paired normal mucosa, advanced adenoma, and carcinoma tissues from six patients with colorectal cancer and adenomas, followed by validation in paired tissues from 30 patients, identified GTP as downregulated throughout disease progression, with GPT mRNA and protein expression being lower in advanced adenoma samples than in adjacent normal mucosa samples, and even lower in carcinoma samples. ...
